Numerous research reports have indicated the coumarin nucleus as a potential candidate for development of anti-inflammatory drugs. Various phytoconstituents such as umbelliferone, scopoletin, columbiatnetin, visniadin, marmin, and many more derived from coumarin nucleus are found to have potent anti-inflammatory as well as antioxidant activities. A large number of coumarin derivatives have also been designed, synthesized, and evaluated to have mild-to-very potent anti-inflammatory activity through different mechanisms. However, despite the continuing efforts in search of these drugs, no major breakthrough has been achieved so far. In the present review, a critical analysis of various reports on naturally as well as the synthetically derived coumarin derivatives having anti-inflammatory activity has been carried out and a structural–activity relationship around the coumarin nucleus has been proposed to assist the medicinal chemists in rationally designing the anti-inflammatory drugs. read more read less

Due to their special structural features and electron-rich environment, imidazole- and benzimidazole-containing drugs bind to a variety of therapeutic targets, thereby exhibiting a broad spectrum of bioactivities. Numerous imidazole- and benzimidazole-based drugs have been extensively used in the clinic to treat various types of diseases with high therapeutic potential. Due to their enormous medicinal value, the research and development of imidazole- and benzimidazole-containing drugs is an increasingly active and attractive topic of medicinal chemistry. This review enlightens the landscape of the discovery and development of imidazole- and benzimidazole-based drugs with an emphasis on structure activity relationship features (SAR), medicinal chemistry, and their mechanism of action. Furthermore, the present review also provides the recent advances in the development of imidazole- and benzimidazole-based drugs along with new perspectives. Despite being an important medicinal moiety, a detailed review on the biologically active potential of indolizine derivative is unavailable. To the best of our knowledge, previous reviews on indolizine derivatives mainly focused on their chemistry and synthesis. The present review discusses the versatile nature of indolizine derivatives and their possible mechanism of action. Important SAR points are discussed with each study to highlight the rationale behind the study.
