Fertilisers, especially nitrogen (N) and phosphorus (P) supplies, are frequently used in agricultural soil management to attain high crop yields. However, the intensive application of these chemical inputs can decrease the quality of agricultural soils and increase the probability of environmental pollution. In this study, the impact of P fertilisation on the diversity of the soil bacterial community was assessed. For this, a culture-independent approach targeting 16 rRNA and phoD genes was used on DNA extracted from pasture soils subjected to three different P fertilisation regimes for a long-term (42 years). As alkaline phosphomonoesterase (ALP) is necessary for mineralisation of organic P, an inverse relationship between the level of potential ALP activity and soil available P was expected. Indeed, a lower ALP activity was observed in soil subjected to higher chemical P fertiliser input. Analysis of the prevalence of three divergent families of ALP (PhoA, PhoD and PhoX) in metagenomic datasets revealed that PhoD is the most frequent ALP in soil samples and was selected as the most representative ALP possessed by the soil bacterial communities. Diversity of the phoD phosphorus mineraliser group, as well as the total bacterial community, was both increased in response to long-term P fertilisation. Specifically, phosphorus fertilisation decreased the relative abundance of certain taxa, including Acidobacteria and Pseudomonas fluorescens. In conclusion, this study shows that P fertilisation affects the microbial diversity of soil ecosystems, which might potentially modulate the soil biogeochemical cycle.

Long-term phosphorus fertilisation increased the diversity of the total bacterial community and the phoD phosphorus mineraliser group in pasture soils

Bacterivores, mostly represented by protists and nematodes, are a key component of soil biodiversity involved in soil fertility and plant productivity. In the current context of global change and soil biodiversity erosion, it becomes urgent to suitably recognize and quantify their ecological importance in ecosystem functioning. Using meta-analysis tools, we aimed at providing a quantitative synthesis of the ecological importance of soil bacterivores on ecosystem functions. We also intended to produce an overview of the ecological factors that are expected to drive the magnitude of bacterivore effects on ecosystem functions. Bacterivores in soil contributed significantly to numerous key ecosystem functions. We propose a new theoretical framework based on ecological stoichiometry stressing the role of C:N:P ratios in soil, microbial and plant biomass as important parameters driving bacterivore-effects on soil N and P availability for plants, immobilization of N and P in the bacterial biomass, and plant responses in nutrition and growth.

Linkages of C:N:P stoichiometry and bacterial community in soil following afforestation of former farmland

Nutrient availability greatly regulates soil microbial processes and functions in tropical forests. However, few studies have explored the impacts of nitrogen (N) addition (100 kg P ha−1 year−1), phosphorus (P) addition (100 kg N ha−1 year−1), and N × P interaction on soil microbial biomass and microbial community composition in tropical forests. We established a field nutrient manipulation experiment in a secondary tropical forest of South China. Soil physicochemical properties and microbial community composition were measured. Analysis of phospholipid fatty acids (PLFAs) was used to determine soil microbial biomass and composition, and both were related to environmental factors by the redundancy analysis (RDA) and principal response curves (PRC). We demonstrated that N addition usually did not affect microbial biomass, which was increased by P addition over 3 years of fertilization. Nitrogen addition decreased soil bacterial biomass but did not affect soil fungal biomass after 3 years of fertilization. After P addition, soil fungal biomass increased faster than soil bacterial biomass, indicating a more sensitive response of soil fungi to P addition than bacteria. Phosphorus addition increased fungi/bacteria ratio (F/B) ratios after 3 years of fertilization. Both N and P additions had different effects on soil microbial community in this tropical forest and, thus, probably altered ecosystem functioning.

Effects of nitrogen and phosphorus addition on soil microbial community in a secondary tropical forest of China

Differential responses of soil bacterial communities to long-term N and P inputs in a semi-arid steppe

The availability of essential nutrients, such as nitrogen (N) and phosphorus (P), can feedback on soil carbon (C) and the soil microbial biomass. Natural cycles can be supplemented by agricultural fertiliser addition, and we determined whether the stoichiometry and nutrient limitation of the microbial biomass could be affected by an unbalanced nutrient supply. Samples were taken from a long-term trial (in effect since 1968) with annual applications of 0, 15 and 30 kg P ha−1 with constant N and potassium. Soil and microbial biomass CNP contents were measured and nutrient limitation assessed by substrate-induced respiration. Linear regression and discriminant analyses were used to identify the variables explaining nutrient limitation. Soil and biomass CNP increased with increasing P fertiliser, and there was a significant, positive, correlation between microbial biomass P and biomass C, apart from at the highest level of P fertilisation when the microbial biomass was over-saturated with P. The molar ratios of C:N:P in the microbial biomass remained constant (homeostatic) despite large changes in the soil nutrient ratios. Microbial growth was generally limited by C and N, except in soil with no added P when C and P were the main limiting nutrients. C, N and P, however, did not explain all the growth limitation on the soils with no added P. Increased soil C and N were probably due to increased net primary production. Our results confirm that C:N:P ratios within the microbial biomass were constrained (i.e. homeostatic) under near optimum soil conditions. Soils with no added P were characterised by strong microbial P limitation and soils under high P by over-saturation of microorganisms with P. Relative changes in biomass C:P can be indicative of nutrient limitation within a site.
