Total phenolics, flavonoids and antioxidant activity of tropical fruit pineapple

Selection of poplar species with greater Cd tolerance and exploiting the physiological mechanisms involved in Cd tolerance are crucial for application of these species to phyto-remediation. The aim of this study is to investigate variation in Cd tolerance among the six poplar species and its underlying physiological mechanisms. Cuttings of six Populus species were cultivated for 10 weeks before exposure to either 0 or 200 μM CdSO(4) for 20 days. Gas exchange in mature leaves was determined by a portable photosynthesis system. Cd concentrations in tissues were analyzed by a flame atomic absorbance spectrometry. Subsequently, Cd amount per plant, bio-concentration factor (BCF) and translocation factor (T (f)) were calculated. Nonenzymatic compounds and activities of antioxidative enzymes in tissues were analyzed spectrophotometrically. Cd exposure caused decline in photosynthesis in four poplar species including Populus cathayana (zhonghua 1). Among the six species, P. cathayana (zhonghua 1) displayed the highest Cd concentrations in tissues, the largest Cd amount in aerial parts, the highest BCF in aerial parts and T (f) under Cd exposure. Under Cd stress, increases in total soluble sugars in roots but decreases in starch in roots, wood, and leaves of P. cathayana (zhonghua 1) were found. Induced O (2) (•-) and H(2)O(2) production in roots and leaves, and increases in free proline, soluble phenolics, and activities of antioxidative enzymes were observed in P. cathayana (zhonghua 1). Based on results of this pot experiment, it is concluded that P. cathayana (zhonghua 1) is superior to other five species for Cd phyto-remediation, and its well-coordinated physiological changes under Cd exposure confer the great Cd tolerance of this species.

Plants exposed to biotic and abiotic stresses generate more reactive oxygen species (ROS) than their capacity to scavenge them. Biological molecules are susceptible to attack by ROS, including several proteins, polyunsaturated fatty acids and nucleic acids. The cellular arsenal for scavenging ROS and toxic organic radicals include ascorbate, glutathione, tocopherol, carotenoids, polyphenols, alkaloids and other compounds. Enzymatic antioxidants including superoxide dismutase, peroxidase, catalase and glutathione reductase detoxify either by quenching toxic compounds or regenerating antioxidants involving reducing power. Various aspects relating to sensors for ROS and signaling role of ROS in plants, improvement of antioxidant systems in transgenic plants and functional genomics approaches used to unravel the reactive oxygen gene network has been discussed.

Reactive Oxygen Species and Antioxidants in Plants: An Overview

The accumulation of capsaicin in different parts of fruit viz, placenta, pericarp and seeds of Capsicum annuum L cv Punjab Lal was compared with the activities of first four enzymes of capsaicin biosynthetic pathway at various physiological stages. Capsaicin accumulation (mg g−1 DW) was about ten fold higher in placenta (63.96), than in pericarp (7.12) and seeds (5.06) in ripe fruits. Capsaicin accumulation was 5.79 mg g−1 DW at 28 DAF in whole fruit. The specific activity of PAL was also ten times higher in placenta, whereas the specific activities of Ca4H, Ca3H and CaOMT were about two times higher in placenta than in other parts of fruit. The trend CaOMT > PAL > Ca4H > Ca3H was observed with peak activity at 28 DAF for Ca3H and CaOMT and at 35 DAF for PAL and Ca4H in placenta. These four enzymes showed low activity during the period up to 21 DAF, and peak activities for these enzymes were obtatined at the time of maximum growth of fruit in length and thereafter.

Accumulation of Capsaicin in Seed, Pericarp and Placenta of Capsicum annuum L Fruit

The biochemical composition of leaf gel isolated from vegetative and micropropagated Aloe vera plants at different stages of development had been compared. The total sugars, fructose, sucrose and starch were higher in micropropagated leaf gel at all developmental stages. The micropropagated plants have higher α-amylase activity at all stages with maximum at 6 month old. Catalase and peroxidase activities were higher in younger leaves (up to 4 month old) in both types of plants. The calcium and magnesium level increases up to maturity and ranges from 0.19 to 1.12% and 0.02 to 0.10% respectively, in conventionally grown plants whereas it was a little higher in micropropagated plants. Micropropagated plants had 12.5% higher phosphorus levels. The micropropagated plants (1 month old) have 60% higher phenol content as compared to conventional plants. The micropropagated mature plants have 55 and 18.6% higher saponin and sterol contents as compared to conventional plants. The composition of aloe vera gel depends upon the growth stages and method of propagation.
