Abstract: Keap1 is a BTB-Kelch substrate adaptor protein that regulates steady-state levels of Nrf2, a bZIP transcription factor, in response to oxidative stress. We have determined the structure of the Kelch domain of Keap1 bound to a 16-mer peptide from Nrf2 containing a highly conserved DxETGE motif. The Nrf2 peptide contains two short antiparallel β-strands connected by two overlapping type I β-turns stabilized by the aspartate and threonine residues. The β-turn region fits into a binding pocket on the top face of the Kelch domain and the glutamate residues form multiple hydrogen bonds with highly conserved residues in Keap1. Mutagenesis experiments confirmed the role of individual amino acids for binding of Nrf2 to Keap1 and for Keap1-mediated repression of Nrf2-dependent gene expression. Our results provide a detailed picture of how a BTB-Kelch substrate adaptor protein binds to its cognate substrate and will enable the rational design of novel chemopreventive agents.

Abstract: The B, R, C1, and Pl genes regulating the maize anthocyanin pigment biosynthetic pathway encode tissue-specific transcriptional activators. B and R are functionally duplicate genes that encode proteins with the basic-helix-loop-helix (b-HLH) motif found in Myc proteins. C1 and Pl encode functionally duplicate proteins with homology to the DNA-binding domain of Myb proteins. A member of the b-HLH family (B or R) and a member of the myb family (C1 or Pl) are both required for anthocyanin pigmentation. Transient assays in maize and yeast were used to analyze the functional domains of the B protein and its interaction with C1. The results of these studies demonstrate that the b-HLH domain of B and most of its carboxyl terminus can be deleted with only a partial loss of B-protein function. In contrast, relatively small deletions within the B amino-terminal-coding sequence resulted in no trans-activation. Analysis of fusion constructs encoding the DNA-binding domain of yeast GAL4 and portions of B failed to reveal a transcriptional activation domain in the B protein. However, an amino-terminal domain of B was found to recruit a transcriptional activation domain by an interaction with C1. Formation of this complex resulted in the activation of a synthetic promoter containing GAL4 recognition sites, demonstrating that this interaction does not require the normal target promoters for B and C1. B and C1 fusions with yeast GAL4 DNA-binding and transcriptional activation domains were also found to interact when synthesized and assayed in yeast. The domains responsible for this interaction map to a region that contains the Myb homologous repeats of the C1 protein and to the amino terminus of the B protein, which does not contain the b-HLH motif. These studies suggest that the regulation of the maize anthocyanin pigmentation pathway involves a direct interaction between members of two distinct classes of transcriptional activators.

Abstract: We present a catalog covering 1.62 deg{sup 2} of the COSMOS/UltraVISTA field with point-spread function (PSF) matched photometry in 30 photometric bands. The catalog covers the wavelength range 0.15-24 {mu}m including the available GALEX, Subaru, Canada-France-Hawaii Telescope, VISTA, and Spitzer data. Catalog sources have been selected from the DR1 UltraVISTA K{sub s} band imaging that reaches a depth of K {sub s,tot} = 23.4 AB (90% completeness). The PSF-matched catalog is generated using position-dependent PSFs ensuring accurate colors across the entire field. Also included is a catalog of photometric redshifts (z {sub phot}) for all galaxies computed with the EAZY code. Comparison with spectroscopy from the zCOSMOS 10k bright sample shows that up to z {approx} 1.5 the z {sub phot} are accurate to {Delta}z/(1 + z) = 0.013, with a catastrophic outlier fraction of only 1.6%. The z {sub phot} also show good agreement with the z {sub phot} from the NEWFIRM Medium Band Survey out to z {approx} 3. A catalog of stellar masses and stellar population parameters for galaxies determined using the FAST spectral energy distribution fitting code is provided for all galaxies. Also included are rest-frame U - V and V - J colors, L {submore » 2800} and L {sub IR}. The UVJ color-color diagram confirms that the galaxy bi-modality is well-established out to z {approx} 2. Star-forming galaxies also obey a star-forming 'main sequence' out to z {approx} 2.5, and this sequence evolves in a manner consistent with previous measurements. The COSMOS/UltraVISTA K{sub s} -selected catalog covers a unique parameter space in both depth, area, and multi-wavelength coverage and promises to be a useful tool for studying the growth of the galaxy population out to z {approx} 3-4.« less
