The Achilles’ heel of senescent cells: from transcriptome to senolytic drugs
Yi-Yi Zhu,Tamara Tchkonia,Tamar Pirtskhalava,Adam C. Gower,Husheng Ding,Nino Giorgadze,Allyson K. Palmer,Yuji Ikeno,Yuji Ikeno,Gene B. Hubbard,Gene B. Hubbard,Marc E. Lenburg,Steven P. O'Hara,Nicholas F. LaRusso,Jordan D. Miller,Carolyn M Roos,Grace C Verzosa,Nathan K. LeBrasseur,Jonathan D. Wren,Joshua N. Farr,Sundeep Khosla,Michael B. Stout,Sara J. McGowan,Heike Fuhrmann-Stroissnigg,Aditi U. Gurkar,Jing-jing Zhao,Debora Colangelo,Akaitz Dorronsoro,Yuan Yuan Ling,Amira S. Barghouthy,Diana C. Navarro,Tokio Sano,Paul D. Robbins,Laura J. Niedernhofer,James L. Kirkland +34 more
TL;DR: The results demonstrate the feasibility of selectively ablating senescent cells and the efficacy of senolytics for alleviating symptoms of frailty and extending healthspan.
Journal ArticleDOI
Chronic senolytic treatment alleviates established vasomotor dysfunction in aged or atherosclerotic mice
Carolyn M Roos,Bin Zhang,Allyson K. Palmer,Mikolaj Ogrodnik,Mikolaj Ogrodnik,Tamar Pirtskhalava,Nassir M. Thalji,Michael A Hagler,Diana Jurk,Leslie A. Smith,Grace Casaclang-Verzosa,Yi Zhu,Marissa J. Schafer,Tamara Tchkonia,James L. Kirkland,Jordan D. Miller +15 more
TL;DR: This is the first study to demonstrate that chronic clearance of senescent cells improves established vascular phenotypes associated with aging and chronic hypercholesterolemia, and may be a viable therapeutic intervention to reduce morbidity and mortality from cardiovascular diseases.
Journal ArticleDOI
Length‐independent telomere damage drives post‐mitotic cardiomyocyte senescence
Rhys Anderson,Anthony B. Lagnado,Damien Maggiorani,Anna Walaszczyk,Emily Dookun,James Chapman,Jodie Birch,Hanna Salmonowicz,Mikolaj Ogrodnik,Diana Jurk,Diana Jurk,Carole J. Proctor,Clara Correia-Melo,Stella Victorelli,Edward Fielder,Rolando Berlinguer-Palmini,A Owens,Laura C. Greaves,Kathy L Kolsky,Angelo Parini,Victorine Douin-Echinard,Nathan K. LeBrasseur,Helen M. Arthur,Simon Tual-Chalot,Marissa J. Schafer,Carolyn M Roos,Jordan D. Miller,Neil Robertson,Jelena Mann,Peter D. Adams,Peter D. Adams,Tamara Tchkonia,James L. Kirkland,Jeanne Mialet-Perez,Gavin D. Richardson,João F. Passos,João F. Passos +36 more
TL;DR: A mechanism by which senescence can occur and contribute to age‐related myocardial dysfunction and in the wider setting to ageing in post‐mitotic tissues is described.
Journal ArticleDOI
TGF-β signalling and reactive oxygen species drive fibrosis and matrix remodelling in myxomatous mitral valves
Michael A Hagler,Thomas M. Hadley,Heyu Zhang,Kashish Mehra,Carolyn M Roos,Hartzell V. Schaff,Rakesh M. Suri,Jordan D. Miller +7 more
TL;DR: Activation of canonical TGF-β signalling is a major contributor to fibrosis and matrix remodelling in MMVD, and is amplified by increases in oxidative stress.
Journal ArticleDOI
Transcriptional and phenotypic changes in aorta and aortic valve with aging and MnSOD deficiency in mice
TL;DR: Although MnS OD does not result in overt cardiovascular dysfunction with aging, compensatory transcriptional responses to MnSOD deficiency appear to be tissue specific.
