The Achilles’ heel of senescent cells: from transcriptome to senolytic drugs
Yi-Yi Zhu,Tamara Tchkonia,Tamar Pirtskhalava,Adam C. Gower,Husheng Ding,Nino Giorgadze,Allyson K. Palmer,Yuji Ikeno,Yuji Ikeno,Gene B. Hubbard,Gene B. Hubbard,Marc E. Lenburg,Steven P. O'Hara,Nicholas F. LaRusso,Jordan D. Miller,Carolyn M Roos,Grace C Verzosa,Nathan K. LeBrasseur,Jonathan D. Wren,Joshua N. Farr,Sundeep Khosla,Michael B. Stout,Sara J. McGowan,Heike Fuhrmann-Stroissnigg,Aditi U. Gurkar,Jing-jing Zhao,Debora Colangelo,Akaitz Dorronsoro,Yuan Yuan Ling,Amira S. Barghouthy,Diana C. Navarro,Tokio Sano,Paul D. Robbins,Laura J. Niedernhofer,James L. Kirkland +34 more
TL;DR: The results demonstrate the feasibility of selectively ablating senescent cells and the efficacy of senolytics for alleviating symptoms of frailty and extending healthspan.
Journal ArticleDOI
A common genetic variant is associated with adult and childhood obesity.
Alan Herbert,Norman P. Gerry,Matthew B. McQueen,I. M. Heid,Arne Pfeufer,Thomas Illig,H.-Erich Wichmann,H.-Erich Wichmann,Thomas Meitinger,David J. Hunter,David J. Hunter,Frank B. Hu,Frank B. Hu,Graham A. Colditz,Anke Hinney,Johannes Hebebrand,Kerstin Koberwitz,Kerstin Koberwitz,Xiaofeng Zhu,Richard S. Cooper,Kristin G. Ardlie,Helen N. Lyon,Helen N. Lyon,Helen N. Lyon,Joel N. Hirschhorn,Joel N. Hirschhorn,Joel N. Hirschhorn,Nan M. Laird,Marc E. Lenburg,Christoph Lange,Christoph Lange,Michael F. Christman +31 more
TL;DR: A dense whole-genome scan of DNA samples from the Framingham Heart Study participants was used to identify a common genetic variant near the INSIG2 gene associated with obesity, suggesting that common genetic polymorphisms are important determinants of obesity.
Journal ArticleDOI
Nef induces CD4 endocytosis: requirement for a critical dileucine motif in the membrane-proximal CD4 cytoplasmic domain.
Christopher Aiken,Jason A. Konner,Nathaniel R. Landau,Marc E. Lenburg,Marc E. Lenburg,Didier Trono +5 more
TL;DR: It is demonstrated that Nef acts by inducing CD4 endocytosis, resulting in its degradation in lysosomes, and that CD4 down-regulation is strongly enhanced by the association of Nef with cell membranes through myristoylation.
Journal ArticleDOI
Dicer, Drosha, and outcomes in patients with ovarian cancer
William M. Merritt,Yvonne G. Lin,Liz Y. Han,Aparna A. Kamat,Whitney A. Spannuth,Rosemarie Schmandt,Diana L. Urbauer,Len A. Pennacchio,Len A. Pennacchio,Jan Fang Cheng,Jan Fang Cheng,Alpa M. Nick,Michael T. Deavers,Alexandra A. Mourad-Zeidan,Hua Wang,Peter R. Mueller,Marc E. Lenburg,Joe W. Gray,Samuel C. Mok,Michael J. Birrer,Gabriel Lopez-Berestein,Robert L. Coleman,Menashe Bar-Eli,Anil K. Sood +23 more
TL;DR: Levels of Dicer and Drosha mRNA in ovarian-cancer cells have associations with outcomes in patients with ovarian cancer, and functional assays indicated that gene silencing with shRNA, but not siRNA, may be impaired in cells with low Dicer expression.
Journal ArticleDOI
Airway epithelial gene expression in the diagnostic evaluation of smokers with suspect lung cancer
Avrum Spira,Jennifer Beane,Vishal Shah,Katrina Steiling,Gang Liu,Frank Schembri,Sean Gilman,Yves-Martine Dumas,Paul Calner,Paola Sebastiani,Sriram Sridhar,John F. Beamis,Carla Lamb,Timothy Anderson,Norman P. Gerry,Joseph Keane,Marc E. Lenburg,Jerome S. Brody +17 more
TL;DR: Findings indicate that gene expression in cytologically normal large-airway epithelial cells can serve as a lung cancer biomarker, potentially owing to a cancer-specific airway-wide response to cigarette smoke.
