Ratiometric analysis of Acridine Orange staining in the study of acidic organelles and autophagy.

TL;DR: The photochemical basis of Acridine Orange (AO) is described and its usefulness as a probe for autophagy quantification is found and it is found that considering the red-to-green fluorescence intensity ratio of AO gives the best correlation with other autophagic assays.

Single-cell analysis challenges the connection between autophagy and senescence induced by DNA damage

TL;DR: It is demonstrated that acute treatment with temozolomide induces DNA damage, a transitory activation of PRKAA/AMPK-ULK1 and MAPK14/p38 and the sustained inhibition of AKT-MTOR produces a transient induction of autophagy, which is followed by senescence, showing that DNA damage-induced Autophagy acts by suppressing apoptosis.
